Transmission of linezolid-resistant Enterococcus isolates carrying optrA and poxtA genes in slaughterhouses

Introduction The presence of linezolid-resistant enterococci found in animal-derived food has attracted attention for possible transmission to human-derived through the chain. Linezolid-resistant farms have been widely reported, but carrying antimicrobial resistance (AMR) genes poxtA , optrA or cfr (D) slaughterhouse environments not well addressed. Methods Enterococcus was isolated from samples collected two slaughterhouses Hangzhou, and linezolidin-resistant identified by PCR. minimum inhibitory concentration (MIC) determined microbroth dilution method. Finally, whole genome strains more linezoline sequenced using Oxford Nanopore Technology Results Here, 291 were 309 (94.17%). A total 4 -positive 42 based on susceptibility test showed that highest rate florfenicol 97.82% multidrug (MDR) 95.65%. Two carried multiple linezolid genes, among which casseliflavus CQFYY22-063 cocarrying duck cecum, faecium CQFYH22-006 sewage first time. Furthermore, revealed gene strain located Inc18-type plasmid pFYY063-optrA-70K, pFYY063- poxtA-12K. Meanwhile, Rep3-type pFYH006-poxtA-25K, chromosome. Discussion Together, deserves extensive attention, indicating need strengthen monitoring different links production chain within One Health concept.

Clinical enterococcal resistance to linezolid is defined by the presence of the G2576T mutation. We evaluated the incidence of genetically proven linezolid resistance among vancomycin-resistant Enterococcus faecium strains and linezolid consumption for a possible association. A relationship was found (r(2) = 0.73, P = 0.03) and predicts increasing resistance with current trends of linezolid use.
